Translators mark text to look good with Formatted View on their screens, but these do not get converted well into real typesets. Is it possible to make the Formatted View more like the output from PTXprint using custom.sty? For example, set column width with hyphenations, etc.? PTXprint respects custom.sty.

The Paratext text editor is not designed to produce a final-output quality layout. It's designed to give you a general idea of what it will look like. Paratext styling does not have the capability to change the styles as fine-grained as you would need for a final output. That's the reason PTXprint and Publishing Assistant exist. The formatting for final-output should be done in those tools.
